Screening of Myxobacteria Carrying Tubulysin Biosynthetic Genes

نویسندگان

چکیده

Tubulysins are a group of secondary metabolites produced by myxobacteria that inhibit the function eukayotic cytoskeleton. We developed pair PCR primers specifically amplified tubulysin biosynthetic genes. Using these primers, eight out eighty-one strains belonging to Cystobacteraceae family harbored putative genes were screened through analysis. The selected included two Archangium gephyra, Stigmatella sp., Vitiosangium cumulatum, and unidentified myxobacteria. LC-MS analysis culture extracts from revealed A. gephyra KYC4066 A B.

PCR detection of type I polyketide synthase genes in myxobacteria.

The diversity of type I modular polyketide synthase (PKS) was explored by PCR amplification of DNA encoding ketosynthase and acyltransferase domains in myxobacteria. The sequencing of the amplicons revealed that many PKS genes were distantly related to the published sequences. Thus, myxobacteria may be excellent resources for novel and diverse polyketides.

A novel biosynthetic pathway providing precursors for fatty acid biosynthesis and secondary metabolite formation in myxobacteria.

Short chain carboxylic acids are well known as the precursors of fatty acid and polyketide biosynthesis. Iso-fatty acids, which are important for the control of membrane fluidity, are formed from branched chain starter units (isovaleryl-CoA and isobutyryl-CoA), which in turn are derived from the degradation of leucine and valine, respectively. Branched chain carboxylic acids are also employed a...

Origin of Saxitoxin Biosynthetic Genes in Cyanobacteria

BACKGROUND Paralytic shellfish poisoning (PSP) is a potentially fatal syndrome associated with the consumption of shellfish that have accumulated saxitoxin (STX). STX is produced by microscopic marine dinoflagellate algae. Little is known about the origin and spread of saxitoxin genes in these under-studied eukaryotes. Fortuitously, some freshwater cyanobacteria also produce STX, providing an i...

Regulation of Flavonoid Biosynthetic Genes in Germinating

Many higher plants, including Arabidopsis, transiently display purple anthocyanin pigments just after seed germination. We observed that steady state levels of mRNAs encoded by four flavonoid biosynthetic genes, PAL7 (encoding phenylalanine ammonia-lyase l), CHS (encoding chalcone synthase), CHI (encoding chalcone isomerase), and DFR (encoding dihydroflavonol reductase), were temporally regulat...

Functional Screening of Phosphatase-Encoding Genes from Bacterial Sources

Phosphatase (APase) enzymes including phytases have broad applications in diagnostic kits, poultryfeeds, biofertilizers and plant nutrition. Because of high levels of sequence diversity among phosphatases,an efficient functional screening method is a crucial requirement for the isolation of the encodinggenes. This study reports a functional cloning screening method for the iso...
